Bradyrhizobium strain and the 15N natural abundance quantification of biological N2 fixation in soybean Scientia Agricola

In commercial plantations of soybean in both the Southern and the Cerrado regions, contributions from biological nitrogen fixation (BNF) are generally proportionately high. When using the 15N natural abundance technique to quantify BNF inputs, it is essential to determine, with accuracy, the 15N abundance of the N derived from BNF (the 'B' value). This study aimed to determine the effect of four recommended strains of Bradyrhizobium spp. (two B. japonicum and two B. elkanii) on the 'B' value of soybean grown in pots in an open field using an equation based on the determination of δ15N natural abundance in a non-labelled soil, and estimate of the contribution of BNF derived from the use of 15N-isotope dilution in soils enriched with 15N. The process of biological nitrogen fixation (BNF), performed by symbiotic nitrogen fixing bacteria with legume species, commonly known as α and β rhizobia, provides high sustainability for the ecosystems. Its management as a biotechnology is well succeeded for improving crop yields. A remarkable example of this success is the inoculation of Brazilian soybeans with Bradyrhizobium strains. Rhizobia produce a wide diversity of chemical structures of exopolysaccharides (EPS). ABSTRACT: To quantify the BNF contribution to legumes using the 15N natural abundance technique, it is important to know the abundance of 15N of the plants grown entirely dependent on BNF (value ‘B’). The aim of the study was to determine the 15N natural abundance of N2 fixed by different Bradyrhizobium strains in symbiosis with one soybean cultivar. Treatments consisted of soybean plants cultivated with and without inoculation with ten Bradyrhizobium strains, in five replicates planted in Leonard jars in a sand/vermiculite mixture. Plants were harvested after 46 days. The ‘B’ values of the aerial tissue (‘Bs’) ranged from -2.6 to -3.9 ‰. Sesbania virgata stimulates the occurrence of its microsymbiont in soils but does not inhibit microsymbionts of other species Scientia Agricola

The legume species Sesbania virgata establishes a specific and efficient symbiosis with Azorhizobium doebereinerae. Previous studies have shown that A. doebereinerae occurrence correlates to the presence of S. virgata. This work aimed to evaluate the occurrence of A. doebereinerae and of other nitrogen-fixing Leguminosae-nodulating bacteria (NFLNB) in soil samples collected adjacent to and 10 m away from the stems of five S. virgata plants in pasture areas. Symbiotic characteristics of isolates from these NFLNB populations were also studied. S. virgata and the four promiscuous legume species Leucaena leucocephala, Macroptilium atropurpureum, Phaseolus vulgaris and Vigna unguiculata were inoculated with soil samples to trap A. doebereinerae and other NFLNB....
